The Tet-On and Tet-Off expression systems use a tetracycline-controlled transactivator which interacts with a promoter to regulate expression of the gene of interest. Fetal bovine serum often contains the antibiotic Tet and extraneous Tet results in unintended expression leading to erroneous results.
